The Klein lab is located in the Biology Department of the Heinrich-Heine-Universität Düsseldorf, Germany (https://www.biologie.hhu.de). The main topic is the regulation of the activity of the Notch signaling pathway by the endosomal pathway and the function of the ESCRT machinery. We are employing imaging, biochemistry, molecular biology, genetics and cell biological techniques to understand the various roles of the endosomal system in the regulation of the Notch activity in Drosophila and cell culture.

Working area and tasks

The project addresses the role of the endosomal ESCRT machinery in the regulation of activity of the Notch pathway. This machinery is the only known device to pinch off membranes away from the cytosol and is used in many processes, ranging from intraluminal vesicle formation over axon pruning to cytokinesis. It is also used by enveloped viruses, such as Ebola and HIV, to exit the cell.

The aim of the project is to investigate the mechanism mediated by the ESCRT machinery, using molecular, cell biological techniques and Drosophila genetics in combination with determination of the atomic structures of the ESCRT-III complex members by Cryo-EM.

The work is in frame of our Collaborative Research Center, CRC 1208 (https://www.sfb1208.hhu.de).

You will enroll as a member of the Heinrich Research Academies (HeRA, https://www.hera.hhu.de/informationen-fuer/promovierende/strukturierte-promotionsprogramme). As a member of the CRC 1208 you are also included in the MB Train training program.

 

Your profile

The candidate has a Master degree in Natural Sciences, experience in cell and molecular biology and is familiar with the Drosophila model system. Ideally, he has also experience in advanced imaging techniques.
